The structure of platelet-activating factor acetylhydrolase (PAF-AH) isolated from boar seminal plasma and examined using mass spectrometry*

چکیده
Platelet-activating factor – acetylhydrolase (PAF-AH) – of boar seminal plasma is a heterogeneous protein consisting of four polypeptides with molecular weights of 43, 55, 65 and 100 kDa. In 2004 the authors of the current report demonstrated that the N-terminal amino acid sequence of 43 kDa polypeptide is homologous with the amino acid sequences of the IgG-binding proteins and zona pellucida-binding adhesion proteins. In the current report, due to the strongly blocked Nterminal amino groups of 55, 65 and 100 kDa polypeptides, their molecular structure was examined with mass spectrometry. In the PAF-AH complex structure the presence of epididymis-specific α-mannosidase, fibronectin, spermadhesins AWN-1 and PSP-II, and IgG-binding proteins was

Platelet-activating factor acetylhydrolase and transacetylase activities in human aorta and mammary artery*
Platelet-activating factor (PAF), the potent phospholipid mediator of inflammation, is involved in atherosclerosis. Platelet-activating factor-acetylhydrolase (PAF-AH), the enzyme that inactivates PAF bioactivity, possesses both acetylhydrolase and transacetylase activities. In the present study, we measured acetylhydrolase and transacetylase activities in human atherogenic aorta and nonatherog...
متن کاملHuman plasma platelet-activating factor acetylhydrolase binds to all the murine lipoproteins, conferring protection against oxidative stress.
OBJECTIVE Plasma platelet-activating factor (PAF) acetylhydrolase (AH) is an enzyme bound with lipoproteins that degrades not only PAF but also PAF-like oxidized phospholipids that are proposed to promote atherosclerosis. In this study, we investigated the distribution of PAF-AH protein among lipoprotein classes by using adenovirus-mediated gene transfer in mice, and we examined its effects on ...

OBJECTIVE To evaluate platelet-activating factor acetylhydrolase (PAF-AH) activity and its relationship with clinical and demographic variables, metabolic control, apolipoprotein A and B levels and the susceptibility of low-density lipoprotein (LDL) to in vitro oxidation in patients with type 1 diabetes mellitus (DM 1). METHODS Forty two patients with DM 1 (27 females) and 48 control subjects...
متن کاملHuman platelets secrete the plasma type of platelet-activating factor acetylhydrolase primarily associated with microparticles.
OBJECTIVE Platelet-activating factor acetylhydrolase (PAF-AH) expresses a Ca2+-independent phospholipase A2 activity and hydrolyzes platelet-activating factor as well as oxidized phospholipids. Two major types of PAF-AH have been described: the plasma type, which is associated with lipoproteins, and the intracellular type II PAF-AH. METHODS AND RESULTS We investigated the type(s) of PAF-AH ex...
متن کاملRelationship between platelet activating factor acetylhydrolase activity and apolipoprotein B levels in patients with peanut allergy
BACKGROUND Platelet-activating factor (PAF) is a highly potent phospholipid mediator responsible for the life-threatening manifestations of anaphylaxis. PAF acetylhydrolase (PAF-AH) inactivates PAF and protects against severe anaphylaxis whereas deficiency of PAF-AH predisposes to severe or fatal anaphylaxis. Determinants of PAF-AH activity have not been studied in patients with peanut allergy....
